Jan 25, 2021 Like other tumors that arise in midline structures of the brain, most DIPGs are histone H3-mutated tumors that harbor H3K27M mutations. DIPG  Jun 30, 2020 In adults, these tumours are most frequently found in the thalamus and can appear as a solid lesion rather than infiltrating lesion (1). To search for potential therapeutic options for this devastating cancer, Lin et al. performed high-throughput screening on patient-derived glioma cells, followed by validation in cell cultures and mouse models of the disease. A review of patients with midline tumors suggests shunting may be a more robust treatment for hydrocephalus in pediatric and adult patients with diffuse midline gliomas. Charts of 43 consecutive midline tumor patients were reviewed with approval from the Institutional Review Board (IRB) to determine the rate of hydrocephalus and the appropriate neurosurgical interventions. Diffuse midline gliomas (DMG) are rapidly fatal tumors of the midbrain in children, characterized by a diffuse growing pattern and high levels of intrinsic resistance to therapy. The location of these tumors, residing behind the blood-brain barrier (BBB), and the limited knowledge about the biology of these tumors, has hindered the development of effective treatment strategies. Some diffuse midline gliomas have changes in histone -related genes, the most common is h3K27M. Diffuse midline glioma refers to astrocytomas that have certain characteristics and are located along the midline of the brain and body. The majority of these tumors are found in the brainstem (which controls many critical functions like breathing, swallowing, and heart rate), but can also be found in other midline structures like the thalamus and Purpose: Our understanding of diffuse midline glioma (DMG) biology inclusive of diffuse intrinsic pontine glioma has been revolutionized by the discovery of novel mutations on the tails of histone 3, leading to the reclassification in 2016 of 'diffuse midline glioma, H3 K27M-mutant.'. Diffuse midline gliomas are aggressive childhood brain tumors that are difficult to access surgically and therefore universally lethal. To search for potential therapeutic options for this devastating cancer, Lin et al. performed high-throughput screening on patient-derived glioma cells, followed by validation in cell cultures and mouse models of the disease.

The location of these tumors, residing behind the blood-brain barrier (BBB), and the limited knowledge about the biology of these tumors, has hindered the development of effective treatment strategies.

In general, glioma treatment options include surgery, radiation therapy, chemotherapy, targeted therapy and experimental clinical trials. Diffuse intrinsic pontine glioma (DIPG) is an aggressive brain tumor that begins in the brainstem in an area called the pons. The pons is responsible for vital life functions as well as the nerves that control vision, hearing, speech, swallowing, and movement. There are about 200-300 new cases of DIPG every year in the United States. The main issue with DIPG is that most of these tumors are not classified by grade because surgery to obtain tissue by biopsy or to remove the tumor is not safe because of the location of the tumor. When these tumors are biopsied, they are usually grade III or grade IV tumors, which tend to behave very aggressively.
